The first major question to ask is - do you want any depth at all to the square you're rotating around?  If you want to make it into a slightly 3D cube, instead of just a perfectly flat 2D plane, it's best to enter into the 3D DVE.
 
Assuming you want a simpler 2D plane, one solution goes as follows:
 
Load an effects tree as a transition (the additive mix from the transition menu will work, just remove the contents). Apply 2 Transform nodes and 2 DVEs. (I've attached an image of what it will look like)
The first transform node (Scale and Rotate) feeds the other transform node (Flip to Back) and the last DVE. The Scale and Rotate transform is where you set your image scale (if needed) and does a full 0 to 180 X or Y rotation animation, in order to flip an entire image.  The Flip to Back transform node feeds the first DVE (Back DVE) and does a -90 rotation on the same axis, with animation.  The Back DVE does nothing except feed into the background of the last DVE node.  The last DVE simply animates it's opacity from 100 to 0 in one frame (or less) at the midpoint of the animation (when the Scale and Rotate node is at 90 degrees - so, might be easier to set a key there to match it).  When the last DVE's opacity is zero, you'll get the background rotating into view during the rest of the animation.
Of course, you need to plug your first image into the last DVE and the second image into the Back DVE.
And, this should all be realtime.
 
Hopefully that all makes sense.  Of course, you could animate it all in the two DVE nodes - but this way, the Scale and Rotate transform node contains the whole of the rotation animation - so, you don't need to play with the two DVE nodes if you want to tweak it later.  You could also use a Composite node for the combining images and doing the opacity animation, instead of using the background input of the last DVE.
 
And, of course, this is all from a developer who works a lot with trees and transform nodes. :)

What is the best way to build a DVE transition that rotates around and reveals another source (not reversed) on the back side?
