Might as well throw my two cents in as well...

Used Tk back when I was doing Perl and, hindsight being 20/20, I probably won't be doing that anytime soon.

Wx is nice but I must admit I sometimes had issues getting to where I wanted to go with it. It is much more than a UI toolkit and as such makes for very portable apps.

Qt is my favorite. Well rounded framework that has a very consistent API. Signal and Slots is a very intuitive way to connect things and it fits my brain. My vote would go for QT. You'll need PyQt of course and it is a pain that Riverbank have taken the Python 2.4 version of the bindings from their web site. If that is an issue for you, I have the bindings for Python 2.4 and Qt 4.1.4.
